The answer, as is often the case, is "it depends".
What synth? Do just a few keys not work? Do no keys work? Does every 8th key not work? In what way do they not work--no note when pressed? No velocity? No aftertouch?
Most often a dead key is due to a dirty or faulty contact. Some synths use leaf-springs, some use J-wires and a bus bar, some use membrane switches, and the procedure for cleaning/repairing them can vary. Other causes of dead keys include a loose or dirty connector between the keybed and the main board; this often causes every X keys to not work.
Generally, the first thing to do is to open it up and visually inspect the key contacts. But if you post what kind of synth/keyboard you're talking about, we can offer more specific advice regarding your gear.

A quick word of warning - if it uses membrane switches - DO NOT CLEAN WITH ELECTRONICS CLEANER!!!!
I just wrecked a half an octave on my Matrix 6 by spraying Radio Scrap Precision Electronics Cleaner around the rubber membranes.
Distilled water is best.
